马铃薯miR159的克隆及人工表达载体构建/Cloning and Construction of Artificial Expression Vector of Potato MiR159

MicroRNAs (miRNAs)是一类非编码小分子RNA,是体内重要的内源性基因调节因子,主要是在转录后水平调控靶基因的表达。本研究是通过人工microRNA技术(artificial microRNA, 简称amiRNA),与PCR技术克隆得到了701 bp的miR 159片段,然后将获得的miR 159片段通过双酶切后链接到植物表达载体pCPB 121,构建了马铃薯amiR 159表达载体。经PCR和酶切分析鉴定,确认载体构建成功。为后期通过马铃薯的遗传转化研究miR 159 / MYB的调控模式提供了帮助。
MicroRNAs (miRNAs) is a kind of non-coding small molecule RNAs, which is an important endogenous gene regulation factor and plays an important regulatory roles on negatively affecting gene expression at post-transcriptional level. In this study a 701 bp fragment of miR 159 was cloned by artificial miRNA and PCR technology. The expressional vector of amiR 159 was constructed with this fragment (701 bp) of miR 159 by double digestion ligated into the plant expression vector pCPB 121. Both PCR and restriction endonuclease digestion confirmed that the vector was successfully constructed. These findings could be helpful for searching on the regulation mode of miR159/MYB through the genetic transformation of potato.
