寄主和非寄主植物中MMDVd正负链比例的研究

摘 要

引物特异反转录结合实时荧光定量PCR技术对MMDVd在寄主和非寄主植物体内正负链的积累差异进行了研究。根据MMDVd基因组序列,设计了3对反向互补的反转录引物,对桑树和拟南芥中的MMDVd的正负链进行引物特异的反转录,荧光定量PCR技术对纯化后的cDNA进行定量分析。分析表明,桑树中,3组反转录引物的结果都是负链高于正链,为2-9倍,而拟南芥中,正负链的比例不定,显示了较高的随机性。本研究可为进一步的类病毒的侵染机制及其寄主域的研究提供参考。
